Brook Street is one of the principal streets on the Grosvenor Estate in the exclusive central London district of Mayfair. It was developed in the first half of the 18th century and runs from Hanover Square to Grosvenor Square. The continuation from Grosvenor Square to Park Lane is called Upper Brook Street where United States Embassy is located on 1 Grosvenor Square.

Features of Brook Street include the grand hotel Claridge's, at the junction with Davies Street, and Le Gavroche, a famous restaurant. The United States Embassy abuts Upper Brook Street and The Handel House Museum is also on Brook Street at number 25. It was the home of the German born baroque composer George Frideric Handel from 1723 until his death. Handel composed works such as The Messiah, Zadok the Priest and the Music for the Royal Fireworks there.

Jimi Hendrix, another famous former resident, lived next door at number 23 for a time.
